چکیده

GSTs (Glutathione S-transferases) are known to catalyze the nucleophilic attack of sulfhydryl group reduced glutathione (GSH) on electrophilic centers xenobiotic compounds, including insecticides and acaricides. Genome analyses polyphagous spider mite herbivore Tetranychus urticae (two-spotted mite) revealed presence a set 32 genes that code for secreted proteins belonging GST family enzymes. To better understand role these in T. urticae, we have functionally characterized TuGSTd01. Moreover, modeled structure enzyme apo form, as well form with bound inhibitor. We demonstrated this protein is S-transferase can conjugate 1-chloro-2,4-dinitrobenzene (CDNB). tested TuGSTd01 activity range potential substrates such cinnamic acid, cumene hydroperoxide, allyl isothiocyanate; however, was unable process compounds. Using mutagenesis, showed putative active site variants S11A, E66A, S67A, R68A mutants, which were residues predicted interact directly GSH, no measurable activity, required enzymatic There several reports associate some acaricide resistance increased . However, found not able detoxify abamectin; fact, inhibits Ki = 101 μM. Therefore, suggest observed abamectin resistant field populations part compensatory feedback loop. In case, production relatively high concentration GSH cells allow maintain physiological functions despite acaricide.

mechanisms of resistance to abamectin in two-spotted spider mite, tetranychus urticae koch (acari: tetranychidae)

tetranychus urticae koch is an injurious pest of, fruit crops, crop plants as well as ornamental plants worldwide. the high reproductive potential of t. urticae and its fairly short life cycle along with the frequent application of acaricides usually required to maintain t. urticae populations below economic thresholds, facilitates the pest's resistance to acaricides, and consequently cont...

Control of two–spotted spider mite Tetranychus urticae Koch (Acari: Tetranychidae) by some selected chemicals

The experiment was conducted to study the effectiveness of four commonly used chemicals viz., deltamethrin, cypermethrin, malathion and sulphur against two-spotted spider mite (TSSM), Tetranychus urticae Koch on potted bean plants and on excised leaf discs. Mite population remained significantly lower in all treated plots except malathion treatment. The population of T. urticae differed signifi...

Protocols for the delivery of small molecules to the two-spotted spider mite, Tetranychus urticae

The two-spotted spider mite, Tetranychus urticae, is a chelicerate herbivore with an extremely wide host range and an extraordinary ability to develop pesticide resistance. Due to its responsiveness to natural and synthetic xenobiotics, the spider mite is becoming a prime pest herbivore model for studies of the evolution of host range, plant-herbivore interactions and mechanisms of xenobiotic r...
